1. Silicization quality of vacuum sampling vessel
There are many varieties of silicone oil used for the treatment of the inner wall of test tubes, soluble in water and insoluble in water, soluble in organic solutions and insoluble in organic solutions, viscosity is very simple liquid and semi-solid liquid. In the meantime, it is not easy to select the silicone oil suitable for the treatment of the lining of the injection vessel, which has outstanding chemical and physiological lazy. The inner wall of the test tube with better silicification should be very lubricated. After a brief water test, it is possible to distinguish the treatment of the inner wall of the test tube. Put 2 milliliters of water into the test tube and then pour it out. The inner wall of the test tube that has been silicified is completely free of small water droplets remaining on it. It is conceivable, then, that the vacuum tube, without the appearance of blood cells hanging on the wall, has now laid an excellent foundation for the separation of high-quality serum.
2. The effect of increased homogeneity of EDTA-K2 on blood routine results
Because the glass tube has a great danger in the safety performance, it has become a trend to replace the glass tube with special plastic PET test tube. Because PET tube has outstanding hydrophobicity, greatly increased the difficulty of vacuum tube production. It is well known that when liquid EDTA-K2 solution is added to PET tubes, because of the hydrophobicity of PET tubes, after a period of time liquid EDTA-K2 will form block crystals at the bottom of the tubes, orients or other parts of the tubes due to some loss of water, making its solubility greatly decreased. Because large EDTA-K2 crystals cannot fully touch the blood and dissolve in the blood specimen in a short time, platelet aggregation simply occurs, the classification results of white blood cells are not satisfactory, and even some coagulation appears in the blood specimen. As we know, the optimal concentration of EDTA-K2 anticoagulant selected for whole blood cell analysis is 1.5 ~ 2.2mg/1ml whole blood. If the concentration of EDTA-K2 is too low and the anticoagulant strength is not good, the appearance of platelet aggregation will be presented. When the concentration of EDTA-K2 is too high, especially in the presence of large crystals of EDTA-K2, some blood cells are in the hypertonic state, where the cells will be temporarily shortened, which will lead to unsatisfactory results of leukocyte classification. In order to avoid the appearance of crystals formed by water loss, the EDTA-K2 crystals should be very finely and evenly distributed throughout the tube wall for an ideal routine blood vacuum. Once the blood enters the test tube, the evenly distributed EDTA-K2 can be quickly mixed with the blood, thus eliminating the appearance of abnormal blood routine results due to vacuum sampling.
3. Evaluation of the application of separation adhesive and coagulant
Parting gel coagulation tube is the most widely used vacuum tube in hospital, but it is not easy to do a good job of this kind of tube. The key to evaluate the quality of the separation gel coagulation tube is as follows: (1) comparing the same blood sample with the blank tube, whether the content of some ions will be error. (2) Put the collected blood specimen tubes containing separation glue in the water temperature chamber, and adjust the temperature to 38 ~ 39℃. After the blood specimen is full of condensation, centrifuge, and then check whether there is oil drop in the serum. There have been many reports of the presence of such an image because oil droplets can cause fatal blockage to the probes of the biochemical apparatus. (3) eliminate the hemolytic elements formed by incorrect blood collection methods, because of the low quality production process, the residual organic solvent of separation glue or coagulant is the chief evil of damaging blood samples. This damage often shows more hemolysis in the specimen, with particles of separation glue floating in the center of the serum. It is also worth mentioning that including foreign famous vacuum tube companies have declared that the blood coagulation tube or coagulant separation glue is centrifuged after collecting blood samples for half an hour, so as to ensure the accuracy of the results to a large extent.
4. Avoid liquid mildew in the vacuum blood collection tube
Sodium citrate anticoagulant tubes for hemagglutination and erythrocyte sedimentation are produced with reasonably high environmental and water requirements for dissolution. If the process and production environment is not correct, or the purity of the dissolved water is not up to the requirements, then the products produced in the test tube after a period of time sodium citrate solution will have flocculent (mold), and then form the inspection results distortion. Because PET tubes have good hydrophobicity, this kind of tubes are not specially packaged, so it is recommended to be used up within three months.
In addition to the above, heparin tubes and potassium oxalate/sodium fluoride tubes are not technically special because of their relatively small dosage, so they will not be described individually here. It is also worth mentioning that vacuum tubes are affected by altitude, so highland vacuum vessels should be selected in high altitude areas.
